(i) reacting nitromethane with deuterium water under the presence of a base and a phase transfer catalyst to give deuterated nitromethane; and reducing the deuterated nitromethane in an inert solvent to form deuterated methylamine; then optionally reacting the deuterated methylamine with an acid to form METHYL-D3-AMINE HYDROCHLORIDE; or (ii) reacting N-(1,1,1-trideuterated methyl)benzosuccinimide with an acid to form METHYL-D3-AMINE HYDROCHLORIDE.
Methyl-d3-amine hydrochloride can be used as a heavy isotope labeling reagent, paired with the light isotope reagent methylamine hydrochloride, to achieve relative quantitative analysis of intact N-glycopeptides.
