Methyl Green is a DNA stain used as a tracking dye in acid buffers for electrophoresis. It helps differentiate bacteria and is also a tissue stain for nucleoli and mitochondria in tissues. It is used with Bismarck Brown frequently.
Methyl Green powder dye is used in various staining methods in microscopy. It is also used with Pyronine Y dye for one stage DNA (green) and RNA (red) staining method. It is also used as a counterstain with other reactions, such as demonstration of enzymatic activity and for mitochondrial staining according to Altmann. Methyl green solution is a suitable counterstain for chloroacetate esterase, nonspecific esterase, alkaline phosphatase, peroxidase, naphthol AS acetate esterase, and acid phosphatase.
