Definition
ChEBI: A sulfate salt resulting from the reaction of 2 eq. guanadrel with 1 eq. sulfuric acid. A postganglionic adrenergic blocking agent formerly used for the management of hypertension, it has been largely superseded by other drugs less likely to cause orthosta
ic hypotension (dizzy spells on standing up or stretching).
brand name
Hylorel (Pharmacia & Upjohn).
General Description
Guanadrel sulfate, (1,4-dioxaspiro[4.5]dec-2-ylmethyl)guanadine sulfate (Hylorel), issimilar to guanethidine monosulfate in the manner in whichit reduces elevated blood pressure. It acts as a postganglionicadrenergic blocking agent by displacing norepinephrinein adrenergic neuron storage granules, therebypreventing release of the endogenous neurotransmitter onnerve stimulation. Guanadrel sulfate has a much shorterhalf-life (10 hours) than guanethidine monosulfate, whose half-life is measured in days. In the stepped-care approachto hypertension, guanadrel sulfate is usually a step 2 agent.
