Niraparib tosylate hydrate is a sulfonic acid organic compound that can be used as a pharmaceutical intermediate.
Mechanism of action
Niraparib tosylate is an orally active poly (ADP-ribose) polymerase (PARP) inhibitor. By blocking the enzymes responsible for DNA repair, niraparib induces cytotoxicity in cancer cells.
Synthesis
(S)-3-(4-aminophenyl)piperidine was obtained by chiral resolution, then condensed with methyl 3-formyl-2-nitrobenzoate to construct a pyrazole ring in the presence of sodium azide. Niraparib was subsequently synthesized under ammonia and acidic conditions.
