Synthesis
General procedure for the synthesis of tert-butyl (1-benzylpiperidin-4-yl)carbamate from 4-amino-1-benzylpiperidine and di-tert-butyl dicarbonate: to an anhydrous solution of 4-amino-1-benzylpiperidine (1 mL, 933 mg, 4.9 mmol) in dichloromethane (20 mL) was added triethylamine (1 mL, 725 mg, 7.2 mmol) and di-tert-butyl dicarbonate ( 1.23 g, 5.6 mmol). The reaction mixture was stirred overnight at room temperature and then diluted with dichloromethane (20 mL). The organic layer was washed sequentially with saturated aqueous sodium bicarbonate (10 mL) and saturated saline (10 mL), dried over anhydrous magnesium sulfate, and concentrated under reduced pressure to give tert-butyl (1-benzylpiperidin-4-yl)carbamate (1.42 g, quantitative yield) as a white solid.
