Synthesis
General procedure for the synthesis of 4-aminopyridin-2-one from 2-chloro-4-aminopyridine: A mixture of 2-chloro-4-aminopyridine (127 mg, 1.0 mmol) and potassium hydroxide (280.6 mg, 5.0 mmol) in toluene (4.0 mL) was heated in a sealed tube and reacted for 72 hr at 170 °C. Upon completion of the reaction, the mixture was cooled to room temperature and the toluene solvent was subsequently removed by distillation under reduced pressure. The residue was purified by rapid chromatography on silica gel (eluent ratio dichloromethane/methanol/ammonia: 78/20/2) to afford the target product 4-aminopyridin-2-one (107 mg, 97% yield) as a light yellow solid.
