Description
20-(tert-Butoxy)-20-oxoicosanoic acid is a non-cleavable ADC linker used in the synthesis of antibody-drug conjugates (ADCs) and in the synthesis of PROTACs.
Chemical Properties
Eicosanedioic acid mono-tert-butyl ester is a solid at room temperature and pressure, with poor solubility in water but some solubility in alcohol organic solvents. It has certain acidity, and the carboxyl unit at one end of the alkyl chain in its structure can be esterified with alcohol compounds under the action of acid catalyst to obtain the corresponding diester derivatives.
Uses
20-(tert-Butoxy)-20-oxoicosanoic acid is a useful research chemical compound.
Uses
20-(tert-Butoxy)-20-oxoicosanoic acid (CAS 683239-16-9) is a high-purity (99%) long-chain aliphatic coupling agent. Its molecular structure features orthogonal protective groups at both ends: one end contains a reactive free carboxyl group, whilst the other end contains a carboxyl group protected by a tert-butyl ester (-tBu). Under the action of a suitable catalyst, the carboxylic acid can undergo a condensation reaction with the amino group of the lysine (Lys) side chain in a peptide, and is used in the preparation of bioactive molecules.
Synthesis
20-(tert-Butoxy)-20-oxoicosanoic acid can be prepared from eicosanedioic acid by esterification.
