Synthesis
Step b: A mixed solution of methyl 2-bromomethyl-3-methoxybenzoate (997 mg, 3.85 mmol) and ammonium hydroxide (9 mL) in THF (19 mL) was reacted with stirring at room temperature for 4 hours. After completion of the reaction, the reaction mixture was concentrated to dryness under reduced pressure. The resulting residue was purified by fast column chromatography (eluent: hexane/ethyl acetate, 40/60 with 1% methanol) to afford 4-methoxyisoindolin-1-one as a white solid (595 mg, 95% yield).
