Application
2-Hydroxy-5-iodobenzonitrile is an organic intermediate that can be used to prepare tert-butyl 4-((2-(aminomethyl)-4-iodophenoxy)methyl)piperidine-1-carboxylate. 4-((2-(aminomethyl)-4-iodophenoxy)methyl)piperidine-1-carboxylate and related derivatives have wide applications in medicinal chemistry and organic synthesis.
Synthesis
(1)Synthesis of 5-iodo-2-hydroxybenzaldehyde oxime
Add 30g 5-iodo-2-hydroxybenzaldehyde to 270ml ethanol, dropwise add 17g hydroxylamine hydrochloride, room temperature stirring overnight, cooling, add water and ethyl acetate, extraction and partitioning, collection of the organic phase, drying, concentration, 23g 5-iodo-2-hydroxybenzaldehyde oxime was obtained.
(2) Synthesis of 2-hydroxy-5-iodobenzonitrile
Add 23g 5-iodo-2-hydroxybenzaldehyde oxime to 190ml acetic anhydride, heat and reflux stirring for 2 hours, concentrate, pour the residue into ice water, add ethyl acetate to extract the liquid, collect the organic phase, dry, concentrate, and get 16g 2-hydroxy-5-iodobenzonitrile separated by column.
