β-羟基异戊酸性质、用途与生产工艺

概述 β-羟基异戊酸又叫3-羟基-3-甲基丁酸。3-羟基-3-甲基丁酸(也被称为“β-羟基-β-甲基丁酸”，“3-羟基异戊酸”，“HMB”)是必需氨基酸中亮氨酸的代谢产物，具有运动时减轻肌肉组织的损伤、增加肌肉和肌肉力量、提高肌肉组织损伤的恢复能力、改善肌肉合成与分解的平衡、维持肌肉和肌肉力量、减少体脂肪、收紧、提高基础代谢等功能的有益化合物。
制备

向烧瓶中加入浓度为13.3重量％的次氯酸钠水溶液347g(0.620mol)，冷却，一边保持在0℃-10℃一边加入20.0g(0.172mol)双丙酮醇，在0℃-10℃继续搅拌15分钟使双丙酮醇与次氯酸钠反应。

然后将反应物用盐酸60.8g(0.600mol)酸化(pH：1.0)后，浓缩至残留重量为125g(约为浓缩前重量的30％)，用乙酸乙酯(45mL×3次)萃取收集3-羟基-3-甲基丁酸(HMB)游离酸，3次萃取液的混合液浓缩得到HMB游离酸14.9g(以HMB换算为0.126mol)。
生物活性 3-Hydroxyisovaleric acid是正常的存在于人类尿液中的代谢物。

The reduced activity of methylcrotonyl-CoA carboxylase (MCC) leads to an elevated excretion of 3-methylcrotonic acid, the product of its hydration (3-hydroxyisovaleric acid: 3-HIA), and 3-methylcrotonylglycine, formed by conjugation with glycine.
体内研究

In male mice (ICR/Jcl) at 8 weeks of age fed the biotin-deficient diet for 6 weeks, the concentration of urinary 3-Hydroxyisovaleric acid is 114.2 mM creatinine, which is higher than the concentration of 35.2 mM creatinine in mice fed the biotin-deficient diet for 4 weeks.
