在室温条件下,将5-溴-2,4-二氯嘧啶(45.6 g,200 mmol)溶解于二恶烷(400 mL)中,随后向该溶液中加入环戊胺(20.4 g,240 mmol)。将反应混合物在室温下持续搅拌6小时。反应完成后,用乙酸乙酯稀释反应混合物,依次用饱和食盐水洗涤,并经无水硫酸镁干燥。减压浓缩除去溶剂,得到5-溴-2-氯-N-环戊基-4-嘧啶胺,为浅黄色固体(56 g,产率100%),该产物无需进一步纯化即可用于后续反应。产物表征数据如下:1H NMR (500 MHz, DMSO-d6) δ 8.23 (1H, s), 7.37 (1H, d, J = 7.3 Hz), 4.31 (1H, m), 1.92 (2H, m), 1.71 (2H, m), 1.53-1.59 (4H, m) ppm; LCMS-ESI (POS), m/z [M + H]+: 实测值 276.0, 计算值 275.9。
