步骤A:在氮气保护下,将2-氨基-4-氟苯甲酸(4.0g,25.8mmol)溶解于脱气的DMF(50mL)中。依次加入HOBt(4.19g,31mmol)、DIEA(5.4mL,31mmol)、EDCI(5.94g,31mmol)和2N氨的甲醇溶液(18mL,36.1mmol)。将反应混合物在室温下搅拌48小时。反应完成后,在减压下浓缩反应液。残余物通过硅胶柱色谱法纯化,得到2-氨基-4-氟苯甲酰胺,为白色固体(2.89g,产率66%)。1H NMR(300MHz,DMSO-d6)δ6.28(m,1H),6.44(m,1H),6.90(br s,2H),7.08(m,1H),7.58(m,1H),7.71(br d,1H)。
