Orexin A is a neuropeptide primarily responsible for regulating metabolism, physiological rhythms, and sleep debt, thereby determining whether an organism should remain awake or sleep. Composed of 33 amino acids, Orexin A contains two pairs of disulfide bonds, exhibits a stable spatial structure, possesses a prolonged physiological half-life, and demonstrates strong biological activity.

Orexin A (human, mouse, rat) is also a hypothalamic neuropeptide that regulates feeding behavior. When administered to the central nervous system of rats, Orexin A stimulates food intake in a dose-dependent manner. This effect lasts for 4 hours post-injection, as Orexin A appears resistant to peptidases. Orexin A can specifically bind to the G-protein-coupled receptor OX1R (IC50 = 20 nM to displace 50% of radiolabeled orexin A bound to this receptor). Orexin A produced in the amygdala can induce wakefulness and plays a role in triggering emotions in humans.
