(S,R,S)-AHPC-C2-PEG3-BCN is a VHL E3 ligase ligand-linker conjugate containing an (S,R,S)-AHPC-based VHL ligand, a PEG3 linker, and a terminal BCN (bicyclononyne) group. It can be used as a PROTAC/targeted protein degradation (TPD) building block for recruiting the VHL E3 ubiquitin ligase in the design and synthesis of heterobifunctional degraders. The BCN group is a strained alkyne that enables copper-free strain-promoted azide-alkyne cycloaddition (SPAAC) with azide-functionalized molecules, making it useful for click-based conjugation and PROTAC synthesis.
