Pomalidomide-PEG3-azide

Pomalidomide-PEG3-azide (CAS No. 2267306-15-8) is an E3 ligase ligand-linker conjugate containing a pomalidomide-derived cereblon (CRBN) ligand , a flexible PEG3 linker, and a terminal azide group. It is a useful building block for the development of PROTACs (Proteolysis Targeting Chimeras) and other targeted protein degradation molecules.

The pomalidomide moiety enables recruitment of the cereblon (CRBN) E3 ubiquitin ligase , while the PEG3 linker provides a flexible spacer for connecting the E3 ligase ligand to a target-protein ligand. The terminal azide provides a versatile click-chemistry handle that can undergo copper-catalyzed azide–alkyne cycloaddition (CuAAC) with alkynes and strain-promoted azide–alkyne cycloaddition (SPAAC) with strained alkyne such as DBCO and BCN.

Pomalidomide-PEG3-azide can therefore be used as a CRBN E3 ligase recruiter and azide-functionalized PROTAC building block in targeted protein degradation, click chemistry, bioconjugation, and medicinal chemistry research.
