BWA-522 PROTAC

BWA-522 is an orally active PROTAC degrader targeting both full-length androgen receptor ( AR-FL ) and androgen receptor splice variant 7 ( AR-V7 ). BWA-522 antagonizes the N-terminal domain of the androgen receptor (AR-NTD) , leading to degradation of AR-FL and AR-V7 and suppression of downstream AR signaling proteins.

BWA-522 induces apoptosis in cancer cells and demonstrates antitumor activity in an LNCaP xenograft mouse model , where it inhibits tumor growth. BWA-522 can be used as a research tool for investigating androgen receptor signaling, targeted protein degradation, AR-FL/AR-V7 biology, and prostate cancer .
