Thalidomide-PEG4-Propargyl

Thalidomide-O-PEG4-Propargyl is a thalidomide-based CRBN ligand containing a PEG4 linker and a terminal propargyl (alkyne) group. The thalidomide moiety enables recruitment of cereblon (CRBN), while the PEG4 linker provides flexible spacing for conjugation to a target-binding ligand.

The terminal alkyne makes this compound suitable for click chemistry, particularly copper-catalyzed azide–alkyne cycloaddition (CuAAC), allowing conjugation with azide-functionalized molecules. It can serve as a useful PROTAC building block for the development of targeted protein degraders and other cereblon-recruiting molecules.
