名称	Alunacedase alfa
描述	Alunacedase alfa (APN-01) is a soluble angiotensin-converting enzyme 2 (hrsACE2) with antiviral activity that reduces the competitive entry of SARS-CoV-2 into cell membranes bound to ACE2. alunacedase alfa mimics ACE2 in vivo and can be used to study novel coronavirus infections, pulmonary hypertension and acute lung injury. 体外活性	Alunacedase alfa 对 SARS-CoV-2 刺突糖蛋白表现出高亲和力结合，估计亲和力约为 1.2 nM。[2] 方法：化合物为 Alunacedase alfa，设置 0.01~1000 μg/mL 浓度梯度，与 SARS-CoV-2（USA-WA1/2020）37℃孵育 1h，再加入 Vero E6 细胞（MOI=0.001）继续培养 4 天；采用中性红法检测细胞病变效应（CPE）与细胞活力。结果：25 μg/mL Alunacedase alfa 可完全中和 SARS-CoV-2，且对细胞无毒性作用。[3]
体内活性	方法：Alunacedase alfa 剂量 100 μg / 只，每日鼻内给药 1 次，连续给药 5 天；实验模型为 BALB/c 小鼠，经鼻感染 1×10⁵ TCID₅₀小鼠适应株 SARS-CoV-2（maVie16）；检测指标包括体重、体温、生存率、肺组织重量。结果：Alunacedase alfa 可使感染小鼠 100% 存活，显著缓解体重下降与肺损伤，降低肺组织重量。[3] 方法：Alunacedase alfa 设置低（0.019 mg/L）、中（0.038 mg/L）、高（0.075 mg/L）3 个剂量组，每日 2 次雾化给药 60 min，连续给药 14 天；实验模型为比格犬；检测临床体征、体重、临床病理、组织病理及毒代动力学指标。结果：Alunacedase alfa 各剂量组均无明显毒性，无全身或器官特异性损伤，最高剂量 0.075 mg/L 为 NOAEL；药物全身暴露量极低，耐受性优异。[3]
