Background

Signal transducer and activator of transcription 3 (STAT3) is a transcription factor which is a member of a family of proteins that mediate signaling by various cytokines and growth factors. STAT3 transmits signals from cell surface receptors to the nucleus to induce gene transcription. It plays critical roles in cell proliferation, survival, differentiation, inflammation and immune response. STAT3 can be activated by phosphorylation on tyrosine 705 in response to numerous cytokines and growth factors. Constitutive activation of STAT3 is frequently observed in many human malignancies and contributes to oncogenesis, tumor progression, metastasis and drug resistance. STAT3 also participates in mitochondrial functions and regulates metabolic reprogramming in cancer cells.
