Background

Tumor protein p53 (TP53) is a transcription factor that responds to diverse cellular stresses to regulate target genes that induce cell cycle arrest, apoptosis, DNA repair, senescence, angiogenesis, and metabolism. p53 acts as a tumor suppressor and is frequently mutated in a wide variety of human cancers. Under normal conditions, p53 protein is maintained at low steady‑state levels through MDM2‑mediated ubiquitination and proteasomal degradation. Upon DNA damage or oncogenic stress, p53 is stabilized and activated by post‑translational modifications including phosphorylation and acetylation, allowing it to bind specific DNA response elements and initiate transcriptional programs for cell fate control.
