Vosoritide is a cyclic peptide composed of 39 amino acids. It functions by binding to a receptor known as natriuretic peptide receptor B (NPR-B), which reduces the activity of fibroblast growth factor receptor 3 (FGFR3). FGFR3 is a receptor typically activated by proteins called acidic and basic fibroblast growth factors, regulating the growth of cartilage and bone. Vosoritide achieves this effect by inhibiting the development (proliferation and differentiation) of chondrocytes, the cells responsible for producing and maintaining cartilage matrix, which is essential for bone growth. Children with achondroplasia may have one of several possible FGFR3 mutations, leading to constitutive (permanent) activation of this receptor, resulting in reduced overall chondrocyte activity and consequently impaired bone growth.
