Bioactivity

名称	Fusaric acid
描述	Fusaric acid is an orally active multi-target inhibitor capable of inhibiting fibrosis-related signaling pathways such as NF-κB, TGF-β1/SMADs, and PI3K/AKT/mTOR. It also inhibits dopamine β-hydroxylase and reduces endogenous levels of norepinephrine and epinephrine. Fusaric acid activates apoptosis-related proteases such as Caspase-3/7, Caspase-8, and Caspase-9, thereby inducing oxidative stress and apoptosis. It chelates divalent metal cations, damages mitochondrial membrane structure, and exerts effects in improving myocardial fibrosis and alleviating cardiac hypertrophy. Additionally, it is applicable in research on esophageal cancer, liver cancer, and other conditions.
体外活性	方法：HepG2人肝癌细胞以25、50、104、150 µg/mL浓度的 Fusaric acid 处理24小时，分别采用比色法检测5-甲基胞嘧啶(5-mC)含量，qPCR法检测DNMT1、DNMT3A、DNMT3B的mRNA表达水平。结果：Fusaric acid 剂量依赖性降低全局DNA甲基化水平，诱导DNA低甲基化且显著降低DNMT1、DNMT3A、DNMT3B mRNA表达。[1] 方法：H9C2细胞加入Fusaric acid 160 μM与异丙肾上腺素(ISP) 10 μM ，共处理24小时，免疫荧光染色检测细胞大小；qPCR和Western Blot检测ANP、BNP、β-MHC、TRPC6表达水平。结果：Fusaric acid 显著抑制ISP诱导的细胞面积增大，同时下调ANP、BNP、β-MHC和TRPC6表达。[2] 方法：小鼠原代心脏成纤维细胞加入Fusaric acid 0–160 μM，预孵育1小时，然后与ISP 10 μM 共处理24小时，Western Blot检测α-SMA、Collagen I、Collagen III表达水平。结果：Fusaric acid 剂量依赖性抑制ISP诱导的胶原蛋白表达，降低纤维化相关基因表达水平。[2]
体内活性	方法：为研究Fusaric acid对ISP诱导的心肌肥厚的影响，选用C57BL/6小鼠灌胃给药Fusaric acid (50, 100 mg/kg) 7天，然后与 ISP (150 mg/kg/天, 皮下注射)同处理7天。结果：Fusaric acid 减轻ISP诱导的心肌组织损伤、胶原沉积和心肌细胞肥大，Fusaric acid 抑制ISP诱导的TGF-β1表达和SMAD2/3磷酸化与p38、ERK1/2和JNK磷酸化。[2]
