Bioactivity

名称Erasin
描述Erasin is a STAT3 inhibitor (IC50 = 9.7 μM) with higher selectivity for inhibiting STAT3 tyrosine phosphorylation than STAT5 and STAT1. It induces apoptosis in various cancer cell lines (e.g., MDA-MB-231, HCC-827) and is effective against Erlotinib-resistant cancer cells.
体外活性方法： HepG2 和 K562 细胞分别用 Erasin 处理，浓度设为 0、5、10、25、50 和 100 μM，孵育时间为 24 小时，并在处理前预孵育 1–2 小时。随后分别使用 IL-6（刺激 STAT3） 和 IFN-γ（刺激 STAT1） 处理细胞，检测各 STAT 蛋白的酪氨酸磷酸化水平。 结果： Erasin 在 HepG2 细胞中以剂量依赖性方式抑制 IL-6 诱导的 STAT3 Tyr705 磷酸化；对 IFN-γ 诱导的 STAT1 Tyr701 磷酸化也有抑制作用，但程度较低；在 K562 细胞中，STAT5 的酪氨酸磷酸化未被抑制。[1]
