Anvatabart is a clinical-stage, site-specific antibody-drug conjugate (ADC) designed for the treatment of HER2 (ERBB2)-positive solid tumors. It is comprised of a fully human anti-HER2 IgG1κ monoclonal antibody, which is site-specifically conjugated to a proprietary, highly potent microtubule inhibitor payload, AS269. This precise conjugation through non-natural amino acid technology aims to produce a homogeneous ADC with a controlled drug-to-antibody ratio (DAR ≈ 1.8). It is designed to deliver cytotoxic payload directly to HER2-expressing tumor cells with high stability in circulation.
