SPACE peptide is a skin-penetrating peptide composed of 11 amino acids in a cyclic polypeptide form. It enhances the local delivery of large molecular weight hyaluronic acid. When binding to cargoes such as small molecules and proteins, SPACE peptide facilitates their penetration through the stratum corneum into the epidermis and dermis. Additionally, SPACE peptide demonstrates increased permeability to various cell types, including keratinocytes, fibroblasts, and endothelial cells, likely via macropinocytosis. Through a transcellular pathway, SPACE peptide enhances the skin penetration of cyclosporine A, and its co-delivery with keratin and cyclosporine A increases its distribution into keratin-rich keratinocytes
