Bioactivity

名称RG-239
描述RG-239 is an orally active TGR5 agonist with an EC50 of 120 nM, significantly more potent than Betulinic acid (EC50= 1.04 μM). It exhibits much higher selectivity for TGR5 over FXRα. The compound enhances mitochondrial activity in adipocytes and promotes neurite outgrowth at higher concentrations. Additionally, RG-239 inhibits LPS-induced iNOS expression and nitrite production in Raw264.7 cells and microglia. RG-239 is applicable for research related to type 2 diabetes.
体外活性RG-239 (Compound 18 dia 2) (10 μM) 能够提升脂肪细胞中线粒体的功能，表现为提升3T3L1脂肪细胞内细胞色素C氧化酶的活性。在1 μM浓度下，RG-239 (1-10 μM) 诱导的神经突长度为9.5 μm，比Retinoic acid (RA) 的效果低；然而，在10 μM浓度时，中位神经突长度达到37.0 μm，与Deoxycholic acid (DCA) 在NSC-34细胞中的效果相近。RG-239 (2 μM, 24 h) 可以抑制LPS诱导的Raw264.7及小胶质细胞中的iNOS表达和亚硝酸盐生成。
体内活性RG-239 (Compound 18 dia 2) (30 mg/kg/天，口服，3 周) 可降低由 10 周高脂饮食 (脂肪供能占比 60%) 诱导的肥胖小鼠的血浆葡萄糖水平。此外，RG-239 (10 mg/kg/天，口服，2 周) 能下调同样饮食条件下 B6J 小鼠肝脏中炎症基因 Tnfα、IL-6 和 IL-1β 的表达。
