名称Sirtratumab vedotin
描述Sirtratumab vedotin (ASG15-ME) is an ADC. Sirtratumab vedotin (ASG15-ME) conjugates a SLITRK6-specific human γ2 antibody (Igγ2) to the small-molecule microtubule-disrupting agent monomethylauristatin E (MMAE) via a protease-cleavable linker.
体外活性Sirtratumab vedotin(ASG15‑ME)可与 SW780 细胞表面表达的 SLITRK6 蛋白发生高亲和力结合，Kd 为 0.018 nM。在 10 μg/mL 浓度下孵育 1 小时后，Sirtratumab vedotin 与 SLITRK6 结合并发生细胞内化过程。经 120 小时处理，可显著抑制 CHP‑212 细胞的存活能力， IC50 为 0.99 nM [1]。
体内活性Sirtratumab vedotin(ASG15‑ME)在小鼠多种异种移植肿瘤模型中，展现出显著的抗肿瘤活性；其给药方式为静脉注射，给药方案包括 0.25–5 mg/kg 剂量、每周 2 次、持续 2～3 周，或采用单次给药模式 [1]。
