Bioactivity

名称Indole-2-carboxylic acid
描述Indole-2-carboxylic acid is an indole carboxylic acid compound with multi-target biological activity, exhibiting neuroprotective, antiviral, and antioxidant properties. Indole-2-carboxylic acid is a competitive antagonist at the glycine site of the NMDA receptor (Ki = 15 μM). Indole-2-carboxylic acid specifically inhibits the potentiating effect of glycine on NMDA-gated currents, blocking the neurotoxicity caused by excessive NMDA receptor activation, and can be used in research on the treatment of neurodegenerative diseases and cerebral ischemic injury. Indole-2-carboxylic acid is also an HIV-1 integrase inhibitor; it interferes with the integration of viral DNA into the host genome and can be used in the development of anti-HIV drugs.
体外活性方法：大鼠(Long Evans，1–10日龄)皮层或脊髓神经元，加入Indole-2-carboxylic acid (0.01-1000 μM) 和甘氨酸 (10 nM – 40 μM)预孵育，预孵育后施加NMDA(30 μM)，记录稳态电流(最后10–20 s平均)。 结果:Indole-2-carboxylic acid (0.01-1000 μM) 在大鼠皮层/脊髓神经元中竞争性抑制甘氨酸对 NMDA 受体的增强作用，IC50 为 105 μM.[1]
体内活性方法：雄性Sprague-Dawley大鼠侧方流体冲击脑损伤，伤后 15 min 静脉注射Indole-2-carboxylic acid (20 或 50 mg/kg)，行为学评估持续至伤后 2 周，神经化学指标于伤后 48 h 测定。 结果：Indole-2-carboxylic acid显著改善创伤所致认知障碍，伤后 2 周Indole-2-carboxylic acid改善神经运动功能。[2]
