Bioactivity

名称NiCur
描述NiCur is an effective and selective inhibitor of CBP histone acetyltransferase (HAT), with an IC50 of 0.35 μM. NiCur blocks CBP HAT activity, downregulates p53 activation and inhibits apoptosis, while promoting methylation of Lysine 27 on histone H3, inhibiting Caspase 3 activity and PARP cleavage.
体外活性NiCur 在 0.5 - 1 μM 浓度范围内作用于 U2OS 细胞时，能以剂量依赖性方式降低 Dox 诱导的 p53K382ac、p53S15p 及 p53 水平[1]。当浓度为 1.5 μM 时，NiCur 可降低 H3K27ac 水平；同样在 1.5 μM 浓度，NiCur 能够恢复细胞增殖能力；在肠上皮细胞中，NiCur 可下调 Dox 介导的 p53 激活，但不影响 H2A.X S139p 水平。NiCur 能够调节基因调控开关，从而重新编程染色质景观，并可阻断 CBP HAT 活性 [1]。
