名称FITC-Dextran (MW 40000)
描述FITC-Dextran (MW 40000) is a kind of dextran fluorescent probe labeled with fluorescein isothiocyanate (FITC) (Ex=495 nm; Em=525 nm). FITC-Dextran (MW 400000) can be used as a tracer marker to reveal heat shock-induced cell injury and detect the early and late stages of apoptosis. In addition, this probe can be used in the study of cell permeability, including the permeability of the blood-brain barrier and the quantitative analysis of its damage degree.
体外活性细胞标记 [1] 本方法适用于凋亡状态的 HeLa 细胞及人外周血单个核细胞（PBMC），而活的 HeLa 细胞与 PBMC 不会被 FITC-Dextran 染色。 1. 采用 43.5℃孵育 1 h、37℃孵育 8 h 的方式诱导细胞凋亡。 2. 将细胞重悬于 100 μL 培养基中，在 Q-prep 管中加入 10 μL 碘化丙啶（PI）与 10 μL FITC-Dextran (MW 40000)混合，使 PI 与 FITC-Dextran 终浓度分别达到 7.5 μM 和 1.13 μM。 3. 将细胞置于室温、避光环境中孵育 25 min。 4. 取 3 mL 已标记的细胞培养基，以 500 g 离心 10 min。 5. 收集离心后的细胞，加入 1 mL 培养基，通过流式细胞术或荧光显微镜检测（PI：Ex=500 nm，Em=600 nm；FITC-Dextran (MW 40000)：Ex=495 nm，Em=525 nm）。 细胞旁渗透性检测 [4] 1. 在 Transwell 小室的基础培养基中加入终浓度为 0.1 mg/mL 的 FITC-Dextran。 2. 孵育 15 min 后，从 Transwell 插件中收集培养基。 3. 检测荧光信号（Ex=485 nm，Em=538 nm）。 4. 依据荧光强度计算 FITC-Dextran 浓度。 5. 根据浓度结果计算渗透率。
体内活性肠屏障功能测定[5]​ 1.对小鼠进行4小时饥饿处理。​ 2.采用FITC-Dextran (MW 40000)对小鼠实施灌胃操作，灌胃剂量为0.6 mg/g。​ 3.在灌胃后4小时内，检测并记录荧光强度（发射波长Em 520 nm）。
