Bioactivity

名称NPB-1575
描述NPB-1575 is a potent, orally active anti-inflammatory agent capable of crossing the blood-brain barrier. It mitigates neuroinflammation and resists ferroptosis by activating IRS2/Nrf2/NF-κB. NPB-1575 protects against cerebral ischemic injury and improves neurological function outcomes, making it suitable for research in focal ischemic stroke.
体外活性NPB-1575 (1-10 μM, 2 h) 通过IRS2/Akt信号通路激活Nrf2并抑制FOXO1，在LPS刺激的BV2细胞中表现出显著的体外抗炎效应。NPB-1575 (1-10 μM) 通过上调IRS2减少了原代小胶质细胞的炎症反应。在原代神经元和小胶质细胞共培养模型中，NPB-1575 (1-10 μM) 通过上调IRS2在缺血性中风中抑制了铁死亡。
体内活性在 pMCAO 大鼠模型中，NPB-1575 (25 mg/kg, p.o., 术后单次给药) 能通过 IRS2 减少脑缺血引起的神经炎症，并抵抗铁死亡。
