Bioactivity

名称Cystemustine
描述Cystemustine is a DNA inhibitor that induces DNA cross-linking, thereby preventing tumor cell proliferation. It exerts cytotoxic effects by disrupting the cell cycle, inducing cell redifferentiation, and altering phospholipid metabolism. Cystemustine shows high antitumor activity and a short plasma half-life in mice, influenced by the timing of administration. It is applicable in research on various malignancies, including melanoma, glioma, renal cancer, head and neck cancer, and colorectal cancer.
体外活性Cystemustine (100, 200 μM, 2 h) 单独使用时不会引发 DNA 损伤，但与 O6-苄基-N2-乙酰鸟苷 (BNAG) 共同使用时，会显著增加 M3Dau 细胞的 DNA 损伤。Cystemustine (50 μM, 4 h) 对 M4Beu 细胞具有细胞毒性，且与 lGgBZ (一种 O6-alkylguanine-DNA alkyltransferase 抑制剂) 联合使用时效力增强。
体内活性Cystemustine (35 mg/kg, i.v., 单次给药，持续 55 天) 显示出显著的昼夜节律毒性变化，毒性最小出现在光照开始后 15-19 小时，最高毒性出现在光照开始后 7 小时。Cystemustine (15 mg/kg, i.v. 或直接注射到肿瘤内，第 1-19 天给药) 在小鼠黑色素瘤模型中引起细胞形态、细胞周期和黑色素含量的变化。Cystemustine (15 mg/kg, 直接注射到肿瘤内，第 11-18 天给药) 导致小鼠黑色素瘤表现出一种新的磷脂代谢表型。
