Bioactivity

名称MitoTracker Deep Red FM
描述MitoTracker Deep Red FM is a deep red fluorescent dye that selectively accumulates in the mitochondrial matrix, with accumulation dependent on mitochondrial membrane potential (Δψ). The dye forms covalent bonds with mitochondrial proteins by reacting with free thiol groups on cysteine residues. After covalent binding, sensitivity to Δψ changes is reduced, but initial staining remains dependent on Δψ. The MitoTracker Deep Red FM probe is suitable for staining living cells or tissues, labeling mitochondria and indirectly detecting changes in mitochondrial membrane potential. Its maximum excitation/emission wavelengths are 644/665 nm.
体外活性方法：使用MitoTracker Deep Red FM标记原代人皮肤成纤维细胞，并通过落射荧光显微镜和自动化线粒体形态分析，将其与TMRM、MitoTracker Red CMXRos、MitoTracker Orange CM-H2TMRos和MitoTracker Green FM比较；采用FCCP诱导线粒体去极化以评价膜电位依赖性。 结果：在正常膜电位下，各探针均适用于自动化形态分析，但定量结果不能互换。FCCP处理后，线粒体定位对膜电位丧失的敏感性顺序为TMRM >> CM-H2TMRos = CMXRos = MitoTracker Deep Red FM > MitoTracker Green FM，表明MitoTracker Deep Red FM具有中等膜电位敏感性。[1]
