Bioactivity

名称MitoTracker Deep Red FM
描述MitoTracker Deep Red FM is a deep red fluorescent dye for mitochondria that selectively accumulates within the mitochondrial matrix dependent on the mitochondrial membrane potential (Δψ). It covalently binds to mitochondrial proteins by reacting with free thiol groups on cysteine residues. Following covalent binding, sensitivity to Δψ changes diminishes, though initial staining remains Δψ-dependent. The MitoTracker Deep Red FM probe is suitable for staining live cells or tissues, serving to label mitochondria and indirectly detect mitochondrial membrane potential alterations. Its maximum excitation/emission wavelengths are 644/665 nm (Storage: Protect from light).
体外活性MitoTracker Deep Red FM 使用方法： 1.1 储存液的配制：用 DMSO 配制 1 mM MitoTracker Deep Red FM。如用 92 μL DMSO 溶解 50 μg MitoTracker Deep Red FM。（建议分装储存液后，于 -20℃ 或 -80℃ 避光保存。） 1.2 工作液的配制：用预热好的无血清培养基或 PBS 按 1:5000-1:50000 稀释储存液，配制成 20-200 nM 的 MitoTracker Deep Red FM 工作液。（根据实际情况调整工作液浓度，现用现配。） 细胞染色（悬浮细胞）： 2.1 离心收集细胞，加入 PBS 洗涤两次，每次 5 分钟。细胞密度 1×10⁶/mL。 2.2 加入 1 mL MitoTracker Deep Red FM 工作液，室温孵育 15-45 分钟。 2.3 400 g 离心 3-4 分钟，弃去上清。 2.4 用 PBS 洗涤两次，每次 5 分钟。 2.5 用 1 mL 无血清培养基或 PBS 重悬细胞，使用荧光显微镜或流式细胞仪观察。 细胞染色（贴壁细胞）： 3.1 将贴壁细胞培养于无菌盖玻片上。 3.2 从培养基中移出盖玻片，吸除多余培养基。 3.3 加入 100 μL 染料工作液，覆盖细胞，孵育 15-45 分钟。 3.4 吸去染料工作液，用培养基洗 2-3 次，每次 5 分钟后，使用荧光显微镜或流式细胞仪观察。 保存条件：-20℃ 避光保存一年。 注意事项： 1. 建议分装储存液，于 -20℃ 或 -80℃ 避光保存，避免反复冻融；-20℃ 可保存 1 个月，-80℃ 可保存半年。 2. 调整 MitoTracker Deep Red FM 工作液浓度需根据实际情况。 3. 本产品仅用于专业科学研究，不得用于临床诊断、治疗，或食品、药品。 4. 操作时请穿实验服并戴一次性手套。 以上信息均来源于已发表文献，具体实验方案请根据实际研究需求进行适当调整。
