Bioactivity

名称Indusatumab vedotin
描述Indusatumab vedotin is an antibody-drug conjugate (ADC) consisting of an antibody targeting human guanylate cyclase C (GCC) conjugated with the microtubule inhibitor monomethyl auristatin. It can inhibit the proliferation of pancreatic cancer cells, arrest the cell cycle at G2/M phase, and induce apoptosis.
体外活性Indusatumab vedotin 在 0.4–25 μg/mL 浓度下作用 72 小时，可对胰腺癌细胞系产生显著的抗增殖效果。当以 12.5 μg/mL 浓度处理 MIA PaCa-2 与 PANC-1 细胞 24 小时后，可诱导细胞周期阻滞于 G2/M 期，并促进磷酸化组蛋白 H3(P-Histone H3)的表达 [1]。
体内活性Indusatumab vedotin 以 10 mg/kg 剂量静脉注射，每周给药 1 次、连续给药至少 17 天，可显著抑制肿瘤生长速率 [1]。 在携带六种异种移植瘤模型之一的小鼠体内，Indusatumab vedotin 表现出与药物暴露量高度相关的生物学效应 [2]。
