名称Trevogrumab
描述Trevogrumab (REGN-1033) is a fully human monoclonal antibody specifically designed to target and inhibit the activity of growth differentiation factor 8 (GDF8), which is also known as myostatin and functions as a negative regulator of muscle growth;Trevogrumab is currently under extensive research of sarcopenia, a complex condition characterized by progressive and generalized loss of skeletal muscle mass and function often associated with aging, wasting atrophy, various chronic diseases, and significant alterations in dietary and nutritional intake.
体外活性方法: 将人骨骼肌成肌细胞与 trevogrumab 孵育 30 min，随后进行 Western blot 分析。 结果: Trevogrumab 可有效抑制 myostatin 诱导的 Smad2/3 活化，并表现出较高的亲和力和特异性。[1]
体内活性方法: CB17-SCID 和 C57BL/6 小鼠接受 trevogrumab 皮下注射给药，剂量为 10 mg/kg，每周两次，持续 4 周。 结果: Trevogrumab 可使小鼠腓肠肌（GA）和胫骨前肌（TA）的肌肉重量显著增加约 20%，并使肌纤维横截面积增加约 15%。[1] 方法: 雄性 C57BL/6 小鼠在研究开始前饲喂 60% 高脂饮食 27 周。随后于 Day 0、4、7、14 和 21 接受 trevogrumab 皮下注射给药，剂量为 10 mg/kg。整个研究期间持续监测小鼠体重和身体组成。 结果: Trevogrumab 可改善肥胖小鼠的瘦体重/脂肪比例，而联合 semaglutide 治疗则可进一步降低整体体脂含量，且降幅最大。[2]
