描述Human growth hormone-releasing factor acetate is a hypothalamic polypeptide and stimulates GH production and release by binding to the GHRH Receptor (GHRHR) on cells in the anterior pituitary[1].
体外活性GHRHR属于II B类GPCR家族成员，主要通过Gs-adenylate cyclase-cAMP信号通路产生作用。激活II类GPCR的肽激素包括GHRH、秘林、类胰高血糖素肽、胃抑制肽(GIP)、垂体腺苷酸环化酶激活肽、皮质酮释放激素、血管活性肠肽、甲状旁腺激素及降钙素相关肽[1]。GHRH在下丘脑弓状核表达并通过门静脉血管释放，通过激活相应的GHRH受体，直接促进垂体生长激素细胞合成和分泌生长激素[1]。
