名称CIB-L43
描述CIB-L43 is a high affinity and orally available TRBP inhibitor (KD = 4.78 nM), which can effectively inhibit the biosynthesis of oncogenic miR-21, increase the expression of PTEN and Smad7, and block the AKT and TGF-β signaling pathways to inhibit the proliferation and migration of hepatocellular carcinoma (HCC) cells.
体外活性CIB-L43 是一种 2-苯基噻唑-5-羧酸衍生物，能以高亲和力结合 TRBP（KD = 4.78 nM），并有效破坏 TRBP 与 Dicer 的相互作用（IC₅₀ = 2.34 μM）。在肝癌细胞中，CIB-L43 以纳摩尔级效力抑制 miR-21 的生成（EC₅₀ = 0.66 nM），进而上调抑癌因子 PTEN 和 Smad7 的表达，抑制 AKT 和 TGF-β 信号通路，显著降低细胞增殖与迁移能力[1]。
体内活性在肝癌小鼠模型中，CIB-L43 展现出良好的药代动力学特性（口服生物利用度为 53.9%）.
