名称Larsucosterol trimethylamine
描述Larsucosterol trimethylamine (DUR-928 trimethylamine) is a potent liver X receptor (LXR) antagonist that modulates endogenous epigenetics, reduces lipid accumulation in hepatocytes, attenuates lipopolysaccharide (LPS) and TNFα-induced inflammatory responses in macrophages, and alleviates LPS- and acetaminophen (ATMP)-induced multi-organ damage.
体外活性在人类肝细胞 HG 诱导的 MASLD 模型中，Larsucosterol 对 DNMTs 有特异性抑制作用。这导致肝细胞内 1,074 个基因启动子区域中的 5mCpG 转化为 CpG ，导致与关键信号通路（如 MAPK-ERK 、钙-AMPK 和 II 型糖尿病通路）相关的基因上调。[1]
体内活性在小鼠肝切除模型中，larsucosterol 可上调与细胞复制有关的基因的表达，包括 Wt1（Wilms' tumor 1）、PCNA（增殖细胞核抗原）、cMyc（骨髓细胞瘤病癌基因）、细胞周期蛋白A、FoxM1b（叉头盒M1b）和 CDC25b（M期诱导磷酸酶2）、 FoxM1b（叉头盒 M1b）和 CDC25b（M 期诱导磷酸酶 2），同时下调细胞周期停滞基因 Chek2（检查点激酶 2） 和凋亡基因 Apaf1（凋亡肽酶激活因子 1） 的表达。[1]
