名称Vilanterol acetate
描述Vilanterol (GW642444) acetate is a long-acting β2-AR agonist. The pEC50 values for vilanterol acetate are 9.4 for β2-AR, 6.4 for β1-AR, and 6.1 for β3-AR, indicating its selectivity in activating airway β2-AR receptors. This activation elevates cAMP levels, resulting in the relaxation of bronchial smooth muscle. Vilanterol acetate can be used in asthma-related research.
体外活性Vilanterol acetate (Compound 13f) 是一种 β 2 受体激动剂，在转染了人 β 1、β 2 和 β 3 受体的中国仓鼠卵巢细胞 (CHO) 中表现出高效 (β 2 : pEC 50 = 9.4)、高活性 (IA = 0.69) 及高选择性 (pEC 50 差异: β 2 /β 1 = 3.0)。在 5 μM 浓度下，经 30 分钟处理后，Vilanterol acetate 在人肝微粒体实验中快速代谢并失活。其在 MDCKII-MDR1 细胞中的膜渗透性中等 (P_app = 34 nm/s)，这可能会降低其全身吸收。对于 β 2 -AR 的选择性极高，相比 β 2 -AR 和 β 3 -AR 亚型的选择性至少高达 1000 倍。
