Bioactivity

名称Paricalcitol
描述Paricalcitol is a selective vitamin D receptor agonist and a vitamin D analog. Paricalcitol potently inhibits the synthesis and secretion of parathyroid hormone (PTH) and is commonly used in studies of secondary hyperparathyroidism in patients with chronic kidney disease.
动物实验After TAC or sham surgery, a subset of the mice is treated with paricalcitol which activates the VDR, at a final dose of 300 ng/kg/day. Paricalcitol is dissolved in a 95% propylene glycol and 5% ethyl alcohol solution. Mice were intraperitoneally injected with paricalcitol (or vehicle only) three times per week on Monday, Wednesday, and Friday for five consecutive weeks. An established anti-hypertrophic and anti-fibrotic treatment, namely the angiotensin II receptor blocker (ARB) losartan is also included. Previous experiments have shown it is feasible and efficacious to dissolve losartan in the drinking water at a concentration of 30 mg/kg/day; mice are treated for five consecutive weeks. So, in total eight groups are studied. Sham (n=10), TAC (n=10), Sham + losartan (Sham-los, n=10), TAC + losartan (TAC-los, n=10), Sham + paricalcitol (Sham-pari, n=10), TAC + paricalcitol (TAC-pari, n=10), Sham + paricalcitol + losartan (Sham-combi, n=10) and TAC + paricalcitol + losartan (TAC-combi, n=10) [2].
体外活性方法：人脐静脉内皮细胞（HUVEC）使用不同浓度 Paricalcitol（50-500 nM）预处理细胞 24 小时，然后更换培养基并加入 100 µM H₂O₂处理 3 小时，使用 MTT 法检测细胞活力。 结果：在 500 nM 浓度下，Paricalcitol 在损伤后处理能显著提高细胞活力。[1]
体内活性方法：为验证 Paricalcitol 在 CPPD 诱导的急性肾损伤(AKI)中的作用，将 Paricalcitol (0.3 μg/kg) 单剂量腹腔注射给 C57BL/6 小鼠，3 d 后腹腔注射 CPPD (20 mg/kg) 诱导感染性 AKI。 结果：Paricalcitol 预处理则显著提升了 GSH、SOD、GPX4、CAT 和 T-AOC 的水平，增强了肾脏的抗氧化防御。[2] 方法：为研究 Paricalcitol 对内膜增生与狭窄的作用，雌性 Wistar Kyoto 大鼠，股动脉导丝内皮剥脱术(左侧)，右侧假手术对照，术后立即开始腹腔注射给药 Paricalcitol (750 ng/kg) ，隔天一次，持续 2 周。 结果：Paricalcitol 组内膜/中膜比(0.17)和狭窄率(28.3%)有下降趋势，但未达统计学显著性。[3] 方法：雄性 Sprague-Dawley 大鼠，行四血管闭塞术(10 分钟)建立全脑缺血模型，缺血后 5 分钟、1 天、2 天、3 天腹腔注射给药 Paricalcitol (1 μg/kg)，观察至缺血后 4 天。 结果：Paricalcitol 组 4 天生存率 100% (8/8)，对照组 62.5% (5/8)。[4]
