名称Carboxymethyl chitosan
描述Carboxymethyl chitosan is a biochemical reagent used as a biomaterial for life science research and as a sulfonylation reagent for organic synthesis and drug discovery. Carboxymethyl chitosan is applied in biomaterials research focusing on biopolymer modification, drug delivery system modeling, and chemical derivatization strategies in medicinal chemistry-oriented experimental systems.
体外活性方法：设置不同浓度Carboxymethyl chitosan，分别作用于皮肤成纤维细胞、施万细胞、人脐静脉内皮细胞、软骨细胞及多种肿瘤细胞，观测细胞增殖、迁移、活力与凋亡指标。 结果： 1.100 μg/mL 作用 4 天，可促正常皮肤成纤维细胞增殖，抑制瘢痕疙瘩成纤维细胞增殖 [1]。 2.Carboxymethyl chitosan水凝胶促细胞迁移效果优于普通壳聚糖水凝胶 [2]。 3.50~200 μg/mL 处理 8h，提升氧化损伤施万细胞活性，剂量依赖性减少 LDH 释放 [3]。 4.0.5~2.0 mg/mL 作用 24~48h，明显抑制人脐静脉内皮细胞迁移 [4]。 5.50~200 μg/mL 孵育 1h，可剂量依赖性拮抗 IL-1β 引发的兔软骨细胞凋亡 [5]。 6.4.0~5.0 mg/mL 干预 48h，降低多株肿瘤细胞活性 [6]。 7.1.0~5.0 mg/mL 作用 12~24h，剂量依赖性抑制肝癌细胞迁移 [7]。
体内活性方法：构建小鼠 H22 肝癌、Lewis 肿瘤模型，分别按不同剂量与给药频次腹腔、静脉注射Carboxymethyl chitosan，观测肿瘤血管生成、瘤体生长及脏器形态；检测高剂量给药后大鼠血液相关指标变化。 结果：Carboxymethyl chitosan按照75~300mg/kg 腹腔给药可抑制 H22 模型肿瘤血管生成 [4]。20~120mg/kg 静脉给药能阻滞 Lewis 肿瘤生长与转移，提升脾脏指数，改善肝肺组织形态 [6]。Carboxymethyl chitosan按照1350mg/kg 腹腔注射不会干扰大鼠凝血、纤溶及血液流变相关指标 [8]。
