名称Eplontersen
描述Eplontersen is a triantennary N-acetylgalactosamine–conjugated antisense oligonucleotide that selectively inhibits both wild-type and mutant transthyretin mRNA, thereby reducing pathogenic TTR protein production and amyloid fibril formation associated with ATTR amyloidosis affecting cardiac and peripheral nerve tissues.
体内活性在表达人类转甲状腺素蛋白 (hTTR) 的转基因 C57BL/6 小鼠体内研究中，Eplontersen 表现出剂量依赖性的基因沉默活性。以 0.6、2 和 6 mg/kg 的剂量（每周一次，持续 3 周）进行皮下注射 (s.c.) 后，该物质导致 TTR 表达降低。在 6 mg/kg 剂量组中，观察到肝脏 TTR mRNA 水平降至基线的 15%，而到第 17 天，血浆 TTR 蛋白水平降至 21% [3]。
