名称GPR34 receptor antagonist 3
描述Compound 5e, a GPR34 receptor antagonist, exhibits selective inhibition of lysophosphatidylserine-induced ERK1/2 phosphorylation in a dose-dependent manner, demonstrating an IC50 value of 0.680 μM without significant cytotoxicity. Additionally, it displays antisensory activity in a mouse neuropathic pain model [1].
体外活性GPR34受体拮抗剂3（30μM，72小时）在HEK293T、COS-7、BEAS-2B、Muller、LX-2及HUVEC细胞系中表现出无明显毒性[1]。
体内活性GPR34受体拮抗剂3以200 mg/kg剂量，每天通过腹腔注射两次，在急性毒性实验中导致C57BL/6J小鼠死亡。而同样的药物以100 mg/kg剂量，腹腔注射方式同频次使用，在重复毒性实验中则显示小鼠无损伤，证明该剂量安全[1]。此外，在每日两次腹腔注射10或20 mg/kg剂量的GPR34受体拮抗剂3后，能够在小鼠神经性疼痛模型中显著降低机械异常性疼痛的表现[1]。
