Bioactivity

名称Nrf2 activator-21
描述Nrf2 activator-21 is a compound that serves as an Nrf2 activator with both antioxidant and neuroprotective properties. It binds to the Keap1Kelch domain, disrupting the Keap1-Nrf2 interaction and activating antioxidant defense mechanisms. Nrf2 activator-21 decreases apoptosis of hippocampal neurons and reduces caspase-3 activity. It targets cerebral ischemia/reperfusion injury (CIRI) by activating the Nrf2 pathway. The compound improves neurological function, alleviates anxiety-like behavior induced by bilateral carotid artery occlusion (2VO) in rats, and enhances memory. Nrf2 activator-21 is applicable in studies of cerebral ischemia/reperfusion injury.
体外活性Nrf2 activator-21 (Compound 7I) (1-15 μM，24 小时) 能显著提高 H2O2 处理的海马神经元细胞的存活率。Nrf2 activator-21 (2.5 μM，24 小时) 还可以减少海马神经元的凋亡并降低 caspase-3 活性。
体内活性在双血管闭塞 (2VO) 诱导的脑缺血/再灌注损伤研究中，Nrf2 activator-21 (Compound 7I) 以 15 mg/kg 剂量通过腹腔注射单次给药，再灌注后 15 分钟显著改善雄性成年 Wistar 大鼠的神经功能。
