Bioactivity

名称Centaurein
描述Centaurein is a flavonoid compound and an IFN-γ promoter enhancer. It increases the activity of NFAT and NF-κB enhancers. Centaurein significantly elevates IFN-γ expression levels in T cells and NK cells and raises IFN-γ concentration in mouse serum. Centaureidin fully relaxes contractions of rat intact aortic rings. Additionally, Centaurein effectively protects mice against Listeria infection.
体外活性Centaurein (5-100 μg/mL, 24-72 h) 可促进 T 细胞系 Jurkat 细胞和脾细胞中 IFN-γ 的转录，并激活 NFAT 和 NF-κB 增强子的活性。Centaurein (100 μg/mL, 24 h) 在 T 细胞中提升 T-bet 和 IFN-γ 的转录，但对 GATA-3 和 IL-4 没有影响。Centaurein (6 h) 预处理能够将巨噬细胞中李斯特菌的感染率从 46% 降低到 15%，此效应可被 IFN-γ 中和抗体部分逆转（感染率上升至 34%）。Centaureidin (10-100 μM) 能通过浓度依赖的方式完全放松 Norepinephrine (NA) (IC 50 = 16.7 μM) 或高钾浓度 (IC 50 = 16.1 μM) 诱导的完整大鼠主动脉环收缩。
体内活性Centaurein (20 μg/只，腹腔注射，单次剂量) 能提高小鼠血清中的IFN-γ水平，并于注射后24小时达到高峰。此外，Centaurein (20 μg/只，腹腔注射，单次剂量) 通过促进IFN-γ的表达和激活巨噬细胞，对小鼠李斯特菌感染表现出预防和治疗的效果。
