名称xStAx-VHLL TFA
描述xStAx-VHLL TFA is a β-catenin degrader formed by coupling xStAx to VHL ligand, has anti-cancer activity, inhibits Wnt signaling, and can be used in the study of cancer.
体外活性在 HEK293T、HCT116 和 SW480 细胞中，10 μM MSAB 和 50 μM xStAx-VHLL TFA 处理 24 小时后，都能将 β-catenin 蛋白降低到相当的水平。在 LoVo 细胞中，只有 xStAx-VHLL TFA 能够降低 β-catenin 蛋白水平，而 MSAB 则没有影响。[1]
体内活性由于APCmin/+小鼠在大约10周龄时开始发展肠道肿瘤，因此对11周龄的APCmin/+小鼠进行了实验。这些小鼠每隔一天通过腹腔注射30 mg/kg的DMSO溶剂或 xStAx-VHLL TFA，持续14天。结果显示，与治疗前（第0天）或DMSO溶剂处理组相比，xStAx-VHLL TFA 处理显著减少了肿瘤的数量。[1]
