Bioactivity

名称SBI-581
描述SBI-581 is an orally active, selective inhibitor of the serine/threonine kinase TAO3 with an IC₅₀ value of 42 nM and antitumor activity. SBI-581 promotes the accumulation of TKS5α in RAB11-positive vesicles and inhibits the formation of invasive pseudopods, thereby suppressing the invasive and migratory capabilities of tumor cells. The pharmacokinetic profile of SBI-581 following intraperitoneal administration in mice is reasonable.
体外活性方法：C8161.9（人黑色素瘤细胞）加入SBI-581（100 nM），处理2 天，Phalloidin labeling检测肿瘤球体大小。 结果：SBI-581 显著抑制肿瘤球体生长。[1] 方法：ECA109、KYSE150细胞加入SBI-581（1 μmol/L），处理24 h，流式细胞术检测细胞凋亡情况。 结果：SBI-581 可以有效诱导ECA109、KYSE150细胞凋亡。[2]
体内活性方法：为研究SBI-581的体内抗肿瘤效果，构建裸鼠 C8161.9 皮下移植瘤模型，肿瘤形成后开始腹腔注射给药(10 mg/kg/天)，每日一次,持续10 天。 结果：SBI-581 显著抑制肿瘤生长（体积减少约 70%），且对体重无影响。[1] 方法：为研究 SBI-581 与顺铂的联合用药效果，BALB/c 裸鼠皮下接种 ECA109细胞构建肿瘤模型，肿瘤形成后开始腹腔注射给药 SBI-581 (10 mg/kg/天)，顺铂 (3 mg/kg/2 天)，持续14 天。 结果：SBI-581 单药有效抑制肿瘤生长，与顺铂联合用药会有更强的效果。[2]
