Bioactivity

名称GL64
描述GL64 is a selective agonist of ADGRD1 with an EC50 of 3.98 μM. It shows low selectivity for the subtypes ADGRD2, ADGRG5, ADGRG6, CELSR1, CELSR2, CELSR3, and ADGRG4. GL64 activates ADGRD1 by mimicking the satchel sequence. It regulates osteoclast maturation through the cAMP-PKA-NFATC1 pathway. Both in vivo and in vitro, GL64 effectively inhibits osteoclastogenesis and prevents bone loss. This compound is suitable for research into osteoclast-related diseases.
体外活性GL64 (10 μM) 能显著增加过表达 ADGRD1 的 HEK293T 细胞中 CRE-荧光素酶活性超过1.5倍，同时也能提高野生型 MEF 的 CRE-荧光素酶和内源性腺苷 cAMP 水平，但在 Adgrd1 −/− 细胞中无明显作用。GL64 (0-100 μM) 在过表达不同粘附 GPCR 的 HEK293T 细胞（如 ADGRD2、ADGRG5、ADGRG6、CELSR1、CELSR2、CELSR3 和 ADGRG4）中不增强 CRE-荧光素酶活性，对非粘附 GPCR (如 GPR68、NPFFR1、GPR183 和 GPRC5B) 也无激活效果。在经 satchel 肽处理后的 ADGRD1 过表达 HEK293T 细胞中，GL64 表现出对 ADGRD1 的弱激动剂活性 (EC 50 = 16.89 μM)。在雄性野生型骨髓来源的巨噬细胞 (BMMs) 中，GL64 (10 μM，6 天) 能抑制其分化为成熟破骨细胞，但对 Adgrd1 -/- 型 BMM 无影响。GL64 (10 μM，6 天) 降低雄性小鼠破骨细胞成熟过程中 Dc-stamp、Acp5 和 Nfatc1 的 mRNA 表达，GL64 (30 μM) 则提升雄性 BMMs 中内源性 cAMP 水平。进一步地，GL64 (10 μM，2 天) 可减少 BMMs 中 NFATC1 的核定位。
体内活性GL64 (30 mg/kg，腹腔注射，每日一次，4 周) 能有效逆转卵巢切除术 (OVX) 导致的绝经后骨质疏松症小鼠模型中的破骨细胞过度活跃和骨质流失。
