名称W23-1006
描述W23-1006 is a novel, selective, and cell-active covalent inhibitor of ALKBH5 (an m6A RNA demethylase). It functions by binding to the C200 residue of ALKBH5 with an IC50 value of 3.848 μM. W23-1006 demonstrates superior selectivity, being approximately 30-fold and 8-fold more potent against ALKBH5 than against FTO and ALKBH3, respectively. By inhibiting ALKBH5 and increasing global m6A abundance, W23-1006 effectively suppresses proliferation, migration, and colonization in triple-negative breast cancer (TNBC) cells.
体外活性W23-1006 (5 μM) 在三阴性乳腺癌细胞中显著上调全局 m6A 修饰水平，并以剂量依赖性方式 (3.16–31.6 μM) 抑制细胞活力。该化合物还能有效抑制细胞迁移与定植，其机制涉及诱导 G2/M 期阻滞及细胞凋亡 [1]。
体内活性W23-1006 (25 mg/kg, i.p.) 在小鼠异种移植模型中显著抑制肿瘤生长。其吸收迅速(Tmax = 15 min)，半衰期为 0.5–1 小时。在高达 250 mg/kg 剂量下未见明显毒性，提示具有良好的体内耐受性 [1]。
