Bioactivity

名称LFM-A13
描述LFM-A13 is a highly selective inhibitor of the non-receptor tyrosine kinase BTK (Bruton's tyrosine kinase) (IC50 = 2.5 μM). It irreversibly severs the downstream signaling network of the B-cell receptor (BCR) by targeted binding to the BTK kinase catalytic domain, exhibiting excellent anti-leukemic and immunomodulatory effects in models of acute/chronic lymphocytic leukemia and severe autoimmune diseases.
体外活性LFM-A13 (100 uM) 抑制 Epo 诱导的 JAK2、BTK 及其下游 STAT5、ERK1/2 磷酸化，并诱导 PTK1 细胞发生显著的周期阻滞 [1][2]。
体内活性LFM-A13 (10-100 mg/kg) 在乳腺癌小鼠模型中表现出剂量依赖性的抑瘤作用，通过调节细胞存活及凋亡因子缓解肿瘤进展 [3][4]。
